詹姆斯·A·加菲爾德遇刺案（英語：Assassination of James A. Garfield）發生在第20任美國總統詹姆斯·艾布拉姆·加菲爾德上任不久的1881年7月2日，當時他在華盛頓哥倫比亞特區中的遭同黨黨員查爾斯·J·古提奧開槍射擊。
加菲尔德上任之初，继续前任总统拉瑟福德·伯查德·海斯开启的改革官员任命的做法，不采用政治分肥，使不少为共和党竞选有功的人失去担任公职的机会，引起党内纷争，而兇手古提奧在加菲尔德代表共和党竞选总统时曾撰写演讲稿为加菲尔德拉票。加菲尔德最终以微弱优势胜出当选总统，古提奧认为是他写的文章有助於加菲尔德最終獲勝。根据曾經的政党分肥，吉托要求成为美国的驻外大使，但他的要求被多次拒绝。古提奧也曾与其他许多求职者一样，到白宫以及国务院外排队等候，但再次遭到美國國務卿詹姆斯·G·布萊恩拒絕。多次受挫後引發古提奧強烈不滿，並打算殺死加菲爾德作為報復。1881年7月2日，古提奧在朝加菲尔德总统连开两枪。加菲尔德的手臂和后背被擊中。79日後的9月19日，加菲爾德因槍傷引起的併發症在紐澤西州去世。
古提奧被判犯谋杀罪，最終被处以绞刑。
加菲爾德成為美國歷史上第二位死於暗殺的總統（第一位為亞伯拉罕·林肯；第三位為威廉·麥金萊；第四位則為約翰·甘迺迪）；時任副總統切斯特·艾倫·阿瑟接任成為總統，並完成加菲爾德餘下的任期。
