航天飞机是美国航空航天局运作的近地轨道航天器，有一定重复使用空间，正式名称为“航天运输系统”，源自1969年的可回收航天器方案，是唯一获资助的方案项目。航空航天局通过一系列航天飞机任务将哈勃望远镜和多颗人造卫星送上太空，并在近地轨道上开展科学实验，参与国际空间站建设和维护。经过1981年的首轮共四次轨道飞行试验，航天飞机任务于1982年全面启动。

1981至2011年间，美国航空航天局共计执行135项航天飞机任务。所有航天器均从佛罗里达州的肯尼迪航天中心起飞，共计飞行耗时1322天19时21分23秒。是其中轨道飞行时间最长的任务，为时17天15时；最短的仅1分13秒，因挑战者号升空后突然爆炸导致。航天飞机先后九次停靠俄国和平号空间站，造访国际空间站的次数多得多，有37次。所有任务的最高海拔约563公里，在维护哈勃望远镜时达成。共计16个国家的355名宇航员参与飞行任务。78次飞行任务以航天飞机在肯尼迪太空中心降落告终，另有54次在加利福尼亚州爱德华兹空军基地降落，还有一次在新墨西哥州唐娜安娜县白沙。

首艘轨道飞行器“企业号”纯为大气层飞行试验而建，没有轨道飞行能力。用于全面轨道飞行的航天飞机起初共建造四架，分别是哥伦比亚号、挑战者号、发现号和亚特兰提斯号。挑战者号和哥伦比亚号分别毁于1986和2003年事故，致14名宇航员因此丧生。第五架全功能轨道飞行器奋进号1991年建成，用于取代挑战者号。2011年7月21日，亚特兰提斯号完成任务，奋进号航天飞机退役。

飞行编号
美国航天飞机计划正式名航天运输系统（，简称），各航天飞机任务的名称就是编号前加“”。1984年开始，所有航天飞机任务都会分配代号，以为例，数字“41”里“4”代表为项目拨款的联邦财政年度，即1984财年，如“51-”则是指原计划在1985财年拨款的项目，1995财年的第3次任务则会以“151-”命名；第2位数字是指航天飞机的发射地点，1代表肯尼迪航天中心，2代表范登堡空军基地的6号太空发射复合体（不过航天飞行从未在6号太空发射复合体升空），最后面的字母表示计划发射顺序。

挑战者号航天飞机失事后，美国航空航天局恢复采用顺序编号体系，数字以项目启动时开始计算。但同原有编号方式不同的地方在于，这些数字是根据最初的计划来定，并不一定同实际发射顺序相符。数字从26开始，首次任务就是-26，其中的“”代表“再次试飞”（），用来区别之前的任务。后缀在使用两年后取消，最后一次使用是。

取消任务
受风险过高影响，曾有一次航天飞机次轨道试飞任务紧急中止。另外还有多次任务因航天飞机的发展，以及挑战者号和哥伦比亚号失事而取消。

有4次任务在航天飞机已进入轨道后缩短，且缩短时间超过1天，首先是因设备故障导致缩短行程。如果接下来的也需要救援，则救援任务的编号会是-301。-115之后，如果一项任务需要救援，则救援任务的编号会以初使任务之后的任务编号为基准，只是把第一位数字改成3。例如如果需要救援，则会以其后的任务为基准，更名为“”。同时，执行救援任务的就是原计划执行-117任务的航天飞机和宇航员。所有救援任务都由4名宇航员执行，根据被救航天飞机上的人员数量不等，救援任务返航时可能会有10或11人。救援任务约持续11天，但所有应急任务都只停留在计划阶段，从未实际执行过。

美国航空航天局没有像之前的航天飞机任务一样为最后一次任务提供应急救援任务组，而是计划在有需要时通过联盟号宇宙飞船开展一对一救援。
