類澱粉-{}-蛋白)又稱淀粉样-{}-蛋白、淀粉样-{}-物质,簡稱類澱粉(amyloid),是一種不可溶的纖維性蛋白質,其在折叠时以β片层形式聚集成纤维,在组织内可沉积形成不溶性斑块,进而破坏组织和器官的功能,造成阿尔茨海默病等几十种疾病。但是,有的淀粉样物质具有正常的生理功能。
类淀粉纤维在形成的初始是正常具生理功能的单体蛋白质(例如:Aβ肽、转甲状腺素蛋白单体),由于基因突变、代谢异常或环境因素,使得这些前体蛋白开始错误折叠并互相吸引,像搭积木一样形成细长的原纤维;最后在组织沉积时,并非单独沉着,而是形成复合物堆积在细胞间质处,“淀粉样物质”其实是这种蛋白质纤维吸附了黏多糖及某些血清成分后的“复合物”状态。
類澱粉在器官中不正常的堆積,會造成類澱粉沉著症。在許多神經性疾病,如阿茲海默症、帕金森氏症中,都可以觀察到神經系統中出現大量類澱粉蛋白的累積沉澱。許多學者相信它可能導致腦部或其他器官退化或功能障礙。
定義
類澱粉蛋白的名稱來自於早期未成熟的碘染色技術,使得科學家誤以為它是一種澱粉(amylum)。當時科學界一直在爭論它到底是屬於脂肪沉澱,或是碳水化合物沉澱,直到最後,才發現它其實是一種蛋白質。
